脑溢安对脑出血大鼠脑内BDNFm RNA表达的影响

摘　　要：目的 观察脑出血模型大鼠脑内脑源性神经营养因子mRNA（BDNF mRNA）的分布以及中药脑溢安对其影响。方法 通过微量注射器向苍白球内注入Ⅶ型胶原酶0．4u制作脑出血模型，用原住杂交法观察脑出血后2、6、12h和l、4、7d共6个时间点BDNF mRNA的表达变化．以阳性细胞计数作为观察指标。结果 正常组、假手术组大鼠脑内皮质、海马、纹状体等处有BDNFmRNA的表达。脑出血后2h上迷部位BDNFIrIRNA的表达增高，但血肿中心区域未见表达信号，6h后上述部位表达信号继续增强，ld后达高峰，4d后表达开始减弱，7d后其表达水平继续下降但仍高于正常，14d后BDNFmRNA阳性细胞数量厦表达水平降至基础水平。两者的阳性细胞主要表达于神经元，其中在6h、1d,4d3个时间点上，两组阳性细胞计数比较有显著性差异（P〈0.01）。结论 脑溢安可促进脑出血后BDNF mRNA的表达。Objective To observe the distribution of brain- derived neurotrophic factor （BDNF） rnRNA in rats with intracerebral hemorrhage （ICH） and effect of Naoyian （NYA）. Methods The ICH animal models were established by microin- jeering 0.4 u Ⅶ eollagenase into the globus pallidus. We observed the expression variation of BDNF mRNA at 2 h,6 h, 12 h, 1 d,4 d and 7 d after ICH by in situ hybridization technique. And positive cell counting served as the observation index. Results Expression of BDNF mRNA were discovered in cortex, hippecampus, stfiatum of normal controls, sham- operated rats. After 2 h of ICH, expression of BDNF mRNA increased, but no expression signals were found in the hematoma region, and after 6 h, expression in above - mentioned region continuously increased, to reached the peak after 1 d, then decreased after 4 d of ICH injury, and decreased continuely but yet highter than the normal level after 7 d , and positive cells come around to the normal level in cells quantity and expression level after 14 d ICH injury. Both positive cells were mainly expressed in neurons. The BDNF mRNA expression level of NYA group were higher than that of the model group at 6 h, 1 d and 4 d time point （ P 〈 0.001） . Conclusion NYA could up- regulated the expression of BDNF mRNA after ICH injury.
