摘  要:目的探讨由泥蚶制得的一组水溶性多肽(简称泥蚶多肽)的抗肿瘤作用.方法以0.25,0.5和1.0 g·L-1的泥蚶多肽处理体外培养的肿瘤细胞株A549和Ketr-3,测定细胞的增殖活性、蛋白质含量和细胞周期的变化.用100,200和400 mg·kg-1剂量的泥蚶多肽静脉注射荷瘤小鼠,计算对S180和H22肿瘤的抑瘤率以及艾氏腹水瘤(EAC)小鼠的生存时间.结果泥蚶多肽在0.25~1.0 g·L^-1内,对A549和Ketr-3细胞的增殖和细胞蛋白质合成具有明显的抑制作用,对A549和Ketr-3的周期阻滞分别为G2~M期和G0~G1期.泥蚶多肽在100~400 mg·kg^-1内,对小鼠S180和H22的抑瘤率分别达到29.35%~55.43%和26.87%~44.12%,明显延长EAC小鼠的生存时间.结论泥蚶多肽体外对不同肿瘤细胞的抑制作用机制不同,体内对小鼠移植性肿瘤具有显著的抑制作用.OBJECTIVE To investigate the antitumor effects of water-soluble pelypeptides from Area granosa Linnaeus in vitro and in vivo. METHODS After the tumor cell lines of A549 and Ketr-3 were cultured with 0.25, 0.5 and 1. 0 g· L^-1 polypeptides in vitro, the proliferation, protein concentration and cell cycle distribution were evaluated by XTF method. Coomassie brilliant blue staining method and flow cytometry, respectively. Transplanted tmnor models of sarcoma 180, hepatocarcinoma 22 and ehrlich ascites carcinoma (EAC) in mice were used to evaluate the antitmnor effect of the pelypeptides. RESULTS The inhibition inlluences on A549 and Ketr-3 cells proliferation were directly proportional to the pelypeptides concentrations in the range of 0.25- 1.0 g· L^-1. It caused a significant G2 - M, G0- G1 phase rrest in A549 and Ketr-3 veils respectively. With the dosage of 100 - 400 mg· kg^-1, the polypeptides inhibited sarcinoma 180 by 29.35 % - 55.43 %, hepatocarcinoma 22 by 26.87% - 44.12% and significantly prolonged the life span of EAC-bearing mice. CONCLUSION The polypeptides from Arca granosa Linnaeus inhibit significantly difterent kinds of tumors in vitro and in vivo.
