摘  要:目的探讨黄芪丹参复方成分影响胎盘血供的分子机理,为临床有效防治胎盘血供不足所致妊娠并发症提供思路。方法提取分离有效中药黄芪、丹参复方成分,运用NOS(一氧化氮合酶)阻滞剂L-精氨酸甲酯(L-NAME)制作一氧化氮合成阻滞大鼠模型,妊娠18d取胎盘组织,核酸原位杂交技术检测胎盘滋养细胞基质金属蛋白酶9(MMP-9)mRNA表达。结果模型组MMP-9mRNA表达明显增加,明显高于空白组和中药黄芪丹参治疗组,中药黄芪丹参治疗组与空白组比较差异无显著意义(P>0.05)。结论黄芪丹参复方成分可能通过抑制胎盘滋养细胞MMP-9mR-NA表达,阻抑蜕膜崩解、防止胎盘后出血,而有利于胎盘滋养细胞侵入及妊娠维持。Objective To provide an effective prevention and treatment of pregnant complications caused by insufficient placental blood supply, the molecular mechanism of Mongolian milkvetch root - Dan- shen root compound that influencing placental blood supply was investigated. Methods The effective components of Mongolian milkvetch root - Dan- shen root were separated and abstracted. The L- arginine methyl ester (L - NAME) nitrogen oxide synthesis blockade model was established. The placenta tissue samples were examined on the 18th day during pregnancy to inspect the expression of placental nutrient cells' MMP- 9 mRNA by nucleic acid in situ hybridization. Results The expression of MMP- 9 mRNA of the model group was significantly higher than that of the blank group and the Mongolian milkvetch root - Dan - shen root treatment group (P〈0.05), however, the treatment group was similar with the blank group (P〉0.05). Conclusions The components of Mongolian milkvetch root - Dan- shen root compound may restrain the degeneration of decidua and prevent retroplacental blooding and then keep the pregnancy going on through restraining the expression of nutrient cell MMP- 9 mRNA.
