摘 要:目的:建立N-辛基-O-硫酸酯基壳聚糖质量分析方法。方法:采用高效凝胶色谱法测定分子质量,湿法消化后重量法测定磺化度(总硫酸根含量),EDTA返滴定法测定游离硫酸根。结果:3批样品重均分子质量分别为9.7×105,8.9×105,7.4×105;数均分子质量分别为2.0×105,1.8×105,1.6×105;总硫酸根含量约为32%;游离硫酸根含量均小于1%。结论:质量分析指标适宜,结果准确,适用于N-辛基-O-硫酸酯基壳聚糖的质量控制。Objective: To establish quality control method for N-octyl-O-sulfate chitosan. Methods: HPGPC was used for the determination of molecular weight. The degree of sulfonic substitution was determined by gravimetric analysis of total sulfate after digestion with nitric acid and hydroperoxide. The content of free sulfate was measured by reverse titration with EDTA. Results: The weight-average molecular weights of three batches of the substituted chitosan were 9.7×10^5, 8.9 ×10^5 and 7.4×10^5, respectively. The corresponding number-average molecular weights were 2.0×10^5, 1.8×10^5 and 1.6×10^5, respectively. The degree of O- sulfate substitution (total sulfate) was about 32% and the content of free sulfate was less than 1%. Conclusion: The test and assay methods for N-octyl-O-sulfate chitosan are suitable and accurate. They can be used for the quality control of N-octyl-O-sulfate chitosan.
