摘 要:目的本文探讨在液体流向上改变简易平行板流动腔与蠕动泵的相对位置能否实现腔内的正压和负压变化。方法改变简易平行板流动腔与蠕动泵的相对位置用生物信号采集处理系统记录平行板流动腔内的压强变化;将兔骨髓间充质干细胞(BM-MSCs)在上述不同条件下进行流体切应力为0.6dyn/cm2的作用刺激24h,用倒置显微镜观察细胞大体形态与MTT法计量正、负压下细胞的数量。结果本模型能提供波动流和稳定流不同的应力培养条件;兔BM-MSCs在0.6dyn/cm2切应力下,正压流组细胞和负压流组的增殖能力,二者差异无统计学意义,P=0.291>0.05,两组细胞形态结构类似。结论模拟正压和负压流的平行板流动腔可为细胞生物力学研究提供了一种有效的实验模型。Objective To investigate whether a simple parallel chamber system could imitate positive or negative pressure stream. Methods The pressure of the parallel chamber system was recorded by the biology message recording system ; the rabbit BM-MSCs was cultured in the medium expose to the fluids shear stress in vitro for 24 hours. The morphology of the cell was observed under optic microscope ,and investigated the proliferation ratio by using MTT-cell proliferation detection. Results The system could imitate the positive and negative stream, offer pulsatile or steady flow conditions;the positive pressure stream vs negative pressure stream shows no significant difference,P=0. 291. Conclusion Simple parallel chamber system imitating positive or negative pressure stream/could provide a effective model for the research of cellular biomechanics.
