摘 要:建立了用高效液相色谱法同时测定丙酮酸产生菌Toru lop sis g labrata 620中的ATP、ADP、AM P、NAD+和NADH的方法,从而描绘了在整个发酵过程中菌体的能荷变化和氧化-还原态趋势,指出菌体的能荷水平和丙酮酸生物合成密切相关,在菌体的能荷水平降低至0.1时丙酮酸开始大量合成,直至发酵末期能荷水平又重新升高。这为进一步提高丙酮酸的产量、研究Toru lop-sis g labrata中物质与能量代谢的关系奠定了基础。The method of determining ATP (adenosine-5'-triphosphate), ADP (adenosine-5'-dipphos- phate), AMP (adenosine-5'-monophosphate), NAD^+ (nicotinamide adenine dinucleotide) and NADH (nicotinamide adenine dinucleotide reduced) in vivo of pyruvate producing strain, Torulopsis glabrata 620 by high performance liquid chromatography (HPLC) was established and the profiles of both of energy charge and oxidationte-reduction state in vivo throughout the fermentation were studied. Energy charge in vivo plays a very important role in the biosynthesis of pyruvate. Pyruvate is accumulated intensively when the energy charge decreases to 0.1 and at the end of the production. It may represent a valuable tool to improve the production of pyruvate and to study the relationship between materials and energy in the cells.
