摘  要:目的探讨Aβ1-15亚单位疫苗接种对AD转基因模型鼠学习记忆能力的影响。方法将24只AD转基因模型鼠(Tg2576鼠)随机分为Aβ1-15组、Aβ42组和对照组,每组8只。9月龄开始分别皮下接种相应的疫苗,每只鼠共6次(抗原量0.1mg/次),接种时程6个月;对照组皮下注射等量PBS加佐剂。疫苗接种前用Y-迷宫作行为学测试,接种后用Morris水迷宫测作行为学测试。结果疫苗接种前,各组Tg2576鼠的空间学习记忆能力无显著性差异(P>0.05);疫苗接种6个月后,Aβ1-15组,Aβ42组鼠的空间学习记忆能力明显高于对照组鼠(P<0.01);Aβ1-15组鼠与Aβ42组鼠相比差异无显著性(P>0.05)。结论Aβ1-15亚单位疫苗接种和其全肽疫苗一样可以改善Tg2576鼠的学习记忆能力,因而可以替代其全肽疫苗用于AD免疫治疗的进一步研究。Objective To explore the effects of Aβ1-15 peptide vaccine on learning and memory ability of transgenic model mice. Methods Twenty-four Tg2576 mice were randomly divided into three groups: Aβ1-15 group, Aβ42 group, and control group, 8 in each group. The mice were injected with Aβ1-15 peptide vaccine, Aβ42 peptide vaccine, and PBS subcutaneously at 9-month old for 6 months. Y-maze was used for behavior test before immunization, and Morris water maze after immunization. Results There was no significant difference in Y-maze behavior test between the three groups (P〉0.05); After six month immunization, the mice in two experimental groups showed increase in spatial learning and memory, compared with control group (P〈0.01). The results in two experiment groups were no significantly different (P〉0.05). Conclusion These results suggest that immunization with Aβ1-15 has the same effect as that of Aβ42. Aβ1-15 peptide vaccine is worthy of further study instead of Aβ42 peptide vaccine.
