摘  要:目的探讨三羟异黄酮(Genistein)对乳腺癌细胞系MCF-7的生长、细胞周期及诱导细胞凋亡的影响。方法培养MCF-7细胞,在其对数生长期加入三羟异黄酮,通过MTT试验,流式细胞仪技术及Giemsa染色法,观察三羟异黄酮对MCF-7生长的影响。结果(1)MTT试验显示,细胞培养第1天,三羟异黄酮20、40、80、160μmol.L-14个剂量组的吸光度值(该值反映对MCF-7细胞生长的抑制作用)分别为0.460±0.018、0.450±0.020、0.440±0.020、0.385±0.040,明显低于对照组(0.520±0.023,P<0.05),第2、3、4天的变化趋势相同于第1天。(2)三羟异黄酮80、160μmol.L-12个剂量组的细胞周期G2%分别为50.0%、57.5%,而对照组为6.8%。(3)三羟异黄酮10、80μmol.L-12个剂量组晚期细胞凋亡率分别为6.51%、20.03%,而对照组为0.50%。结论三羟异黄酮对MCF-7的生长有明显的抑制作用;能阻滞细胞周期的正常进行,但主要在细胞周期的后期(G2期)发挥作用;能诱导细胞凋亡。Objective To study the effects of genistein on growth inhibition, the cell cycle blocking, cell apoptosis induction of mastopathia cell MCF-7. Methods MCF-7 cells were normally cultured, then during the cell logarithm growth period, the cells were treated with genistein of different doses. The growth of mastopathia cell MCF - 7 was observed by MTT assay, flow cytometer, and giemsa staining. Results ① MTT assay showed that the absorptions (indicating the inhibition on cellular growth) of 20, 40, 80, 160 μmol·L^-1 of genistein were 0.460 ±0.018, 0.450 ± 0.020, 0.440 ± 0.020,0. 385 ± 0.040 respectively on the 1st day of cell culture, which were lower than that of control(0. 520 ± 0.023, P 〈 0.05). Tne changes on the 2nd,3rd,gth day were similar to the 1st day. ②The cell cycle G2% of 80,160 μmol·L^-1 of genistein (50.0 %, 57.5 % respectively) were higher than that of control(6.8 % ). ③The apoptotic rates of 10,80 μmol·L^-1 of genlstein were 6.51% ,20.03% respectively while that of control was 0.50%. Conclusion Genistein can obviously suppress the cell growth, block the cell cycle (especially in G2 period), and induce the cell apoptosis.
