摘 要:目的:观察几种常用化疗药物对人肝癌耐药细胞BEL-7404/ADM端粒酶活性的影响。方法:利用端粒重复扩增实验(TRAP)结合非变性聚丙烯酰胺凝胶电泳银染法,测定BEL-7404/ADM耐药细胞经过多种化疗药物[顺铂(DDP)、阿霉素(ADM)、丝裂霉素(MMC)、长春新碱(VCR)]不同浓度和时间作用后端粒酶活性的变化。结果:当各种药物高浓度处理细胞24 h后再去除药物,DDP对端粒酶的活性完全抑制,MMC有部分抑制,ADM和VCR无抑制作用;低浓度药物处理细胞3 d,其结果同高浓度相似。结论:高浓度DDP和MMCC在短时间内或低浓度长时间作用对人肝癌耐药细胞BEL-7404/ADM端粒酶活性有显著抑制作用,抑制作用可能与药物的作用浓度和时间有关。Objective:To investigate the inhibition of telomerase activity by several chemotherapeutic drugs regularly used in human liver cancer BEL-7404/ADM cells. Methods:By using telomerase repeated amplification protocol (TRAP) combined with PAGE silver staining, we detected the change of telomerase activity of human liver cancer cell line BEL-7404/ADM by DDP, ADM, MMC, and VCR at different concentration (c) and different time periods. Result:In high (c), the telomerase activity of cells was completely inhibited by cisplatin and partly inhibited by mitomycin C when the drug was removed after cells were recultured for 24 hours at high (c) drug treatment. However, doxorubicin and vincristine had no effect. At low (c), 3d after drug treatment the effects were similar. Conclusion:Cisplatin and mitomycin C of high (c) treated for a short time period or low (c) of a long time period can inhibit telomerase activity of human liver cancer BEL-7404/ADM cell , which maybe correlates with its dose and time period of the drug.
