流式细胞术分析乳腺癌前哨淋巴结淋巴细胞亚群的两种方法比较

摘　　要：目的:探讨流式细胞术三色荧光/侧向角散射设门(CD45/SSC-Gating)与散射光设门(FSC/SSC-Gating)两种设门方法对乳腺癌前哨淋巴结淋巴细胞亚群分析的差异及其临床应用价值。方法:将前哨淋巴结制备成单细胞混悬液,用CD45/SSC、FSC/SSC两种方法设门,流式细胞仪直接计数前哨淋巴结中淋巴细胞亚群的百分比,并用统计学方法分析比较结果。结果:FSC/SSC设门组中总淋巴细胞的百分比(TL)为(79·42±5·02)%,CD45/SSC设门组中TL为(97·66±4·60)%(P<0·05)。同时两个设门组的CD3+T,CD4+T,CD8+T测定值比较,差异亦有统计学意义(P<0·05)。结论:在分析乳腺癌前哨淋巴结淋巴细胞亚群时,CD45/SSC设门优于FSC/SSC设门。Objective. To study the differences of two kinds of flow eymetrie methods CD45/SSC-Gating and FSC/SSC-Gating, which were used to analyze the percent of lymphocyte subsets of the sentinel lymph nodes. Methods. Using two kinds of flow cymetric methods CD45/SSC-Gating and FSC/SSC-Gating, the percent of lymphocyte subsets of the sentinel lymph nodes was tested by flow cytometry directly, and the results were analyzed with statistical method. Result： FSC/SSC-Gating was （79.42 ± 5.02）%, CD45/SSC- Gating was （97.66±4.60）%（ P 〈0.05）. And in the two kinds of lymphocyte gate（LG）,the respective percentages of CD3^＋ ,CD4^＋ T,CD8^＋ T were significantly different（ P 〈0.05）. Conclusion： In analyzing the percent of lymphocyte subsets of the sentinel lymph nodes of breast cancer,CD45/SSC-Gating is better than the FSC/SSC-Gating.
