摘 要:目的 为观察并比较不同型流感病毒诱导肿瘤细胞凋亡能力的差异,并探讨肿瘤细胞端粒酶活性变化与凋亡之间的关系。方法 用A1、B型流感病毒及经紫外线照射后的病毒分别感染Hela细胞,于不同时间取细胞分别在电镜下观察细胞的形态,Annexin V-FITC染色流式细胞仪检测凋亡百分率,并用PCR-ELISA法检测肿瘤细胞端粒酶活性。结果 A1型流感病毒(UV-A1)及B型流感病毒(UV-B)作用于Hela细胞后24h,电镜下可见细胞凋亡的特征性改变。流式细胞仪检测肿瘤细胞凋亡百分率随时间延长而增高,于12-24h达高峰,其中UV-B诱导凋亡百分率最高达38.6%,Uv-A1最高为23.9%。紫外线照射后的UV-B最高达到20.9%。UV-A1最高达到10.1%;此外,Hela细胞端粒酶活性均有所下降,其中UV-B诱导后的Hela细胞端粒酶活性下降比A1型更明显,同时,紫外线照射后的UV-B诱导的Hela细胞端粒酶活性也略降低。结论 UV-B在导肿瘤细胞凋亡的能力及降低其端粒酶活性方面都比UV-A1要强,而且经紫外线照射后的UV-B仍然具有一定的诱导Hela细胞凋亡的能力;流感病毒降低肿瘤细胞的端粒酶活性可能是其诱导肿瘤细胞凋亡的又一重要机制。Objective To observe and compare the difference of apoptosis in tumer cells induced by different influenza viruses and investigate its correlation with the variation of telomerase activity. Methods The shapes of Hela cells were observed in different time in the electron microscope after they were infected by influenza virus type A1, B and ultraviolet radiation vires respectively. Annexin V- FITC staining flow cytometry (FCM) was used to observe the percentage of apoptosis and the telomerase activity in turner cells was tested by PCR - ELISA. Results Typical apoptosis in Hela cells were observed in microscope after infected by influenza viruses type A1 and B 24 hours.The percentage of apoptosis increased along with time expanded and culminated in 12 to 24 hours.The maxima of the percentage of apoptosis induced by influenza virus type B, A1, ultraviolet radiation inlfluenza B virus (UV- B) and ultraviolet radiation inlfluenza A1 virus (UV- A1) came to 38.6%, 23.9%, 29.9%and 10.1% respectively. Furthermore, the telomerase activity in Hela cells all decreased, in which the decreasing induced by influenza type uv- B virus was more apparent than influenza A1 virus. At the same time, the telomerase activity in Hela cells induced by ultraviolet radiation inlfluenza B virus also dropped appreciably. Conclusion It comes to the conclusion that influenza B virus is stroner than influenza A1 virus in the ability of inducing apoptosis and decreasing telomerase activity in turner cells. Moreover, the ultraviolet radiation inlfluenza B virus still has the certain ability of inducing apoptosis in Hela cells. It may be is another important mechanism of inducing apoptosis in turner cells that influenza viruses can decreasing the telomerase activity in turner cells.
