摘  要:目的初步探讨生长抑素受体(SSTR)介导90Y-DOTA-Octreotide[90Y-DOTATATE]靶向放疗对裸鼠人肝癌移植模型的抗肿瘤效应。方法合成90Y-DOTATATE,用裸鼠人肝癌移植模型,观察该靶向药物对肿瘤体积、重量、肿瘤细胞细胞器等的影响。结果治疗后90Y-DOTATATE治疗组肿瘤体积为628.32 mm3±403.01 mm3,奥曲肽组肿瘤体积为2073.84±537.61(P<0.01),而生理盐水对照组为2624.91±1257.82 mm3(P<0.01);90Y-DOTATATE组瘤重抑制率为74.67%。细胞超微结构检查发现90Y-DOTATATE治疗组肿瘤细胞损伤主要表现为内质网空泡化、线粒体水肿、细胞核浓缩、染色质边缘化乃至裂解,并可见凋亡小体。结论生长抑素受体介导的90Y-DOTATATE靶向放射治疗确有良好的体内抗肿瘤效应,是一值得进一步深入研究的靶向药物。Objective To elementarily investigate the anti - carcinoma effect of targeted radiotherapy using 90^Y-DOTA-Octreotide [90^Y-DOTATATE] mediated by somatostatin receptor (SSTR). Methods Freshly prepared 90^Y- DOTATATE under the guidance of Medical Isotopes Research Center, Peking University. Male BALB/c nude mice were transplanted with HepG2 hepatocarcinoma cell line mass subcutaneously near right forefoot to establish human hepatocarcinoma model. The mice were divided into 3 groups with 10 mice in each group. Tumor volume and tumor weight was measured, organelle of tumor cells was observed under electron microscope. Results The purity of 90^Y labelled octreotide was more than 95 %. The mean tumor volume of 90^Y- DOTATATE (10 mCi/kg), octreotide and NS on human hepatocarcinoma in nude mice were 628.32 ± 403.01 mm^3 , 2073.84 ± 537.61 mm^3 and 2624.91 ± 1257.82 mm^3 respectively ( P 〈 0.01 ), and the tumor inhibition rate of the former was 74.67 %. Vacuole change of endoplasmic reticulum, swell of metechondria, concentration of nucleus and even apoptosis body could be seen through electron microscope. Conclusion The targeted radiotherapy using 90^Y- DOTATATE mediated by SSTR had a certain extent in vivo anti - carcinoma effect.
