大鼠血管中α_1-肾上腺素受体的同源性减敏不受β-肾上腺素受体和血管内皮影响

摘  要:目的:确定在大鼠血管中β-肾上腺素受体和血管内皮对α_1-肾上腺素受体(α_1-AR)同源性减敏有否影响。方法:采用大鼠离体血管收缩功能实验。结果:经去甲肾上腺素(NE)预温育的主动脉和肾动脉由α_1-AR介导的收缩效应较对照明显降低,而5-羟色胺和氯化钾引起的收缩效应均无明显改变,有或无β-AR激动内皮的存在对上述改变并无明显影响。结论:血管在α_1-AR经激动剂持续作用后产生的减敏为同源性减敏,不受β-肾上腺素受体和血管内皮的影响。Objective:The effects of norepinephrine(NE)pre-incubation on the vasoconstriction induced by adrenoceptor(α1-AR)agonist or 5-HT or KCl and the roles of endothelium and β-AR on the desensi- tization were investigated.Methods:Functional experiments in isolated rat aortae and renal arteries were performed.Results:NE pre-incubation significantly reduced α1-AR mediated vasoconstriction,but did not affect the vasoconstriction induced by 5~HT and KCl.The NE pre- the preparations with or without endothelium and in the presence or ab-sence ofβ-AR agonist during the pre-incubation. Conclusion:The persistent activation of the α1-AR by agonists causes homologous desensitization which was not affected by endothelium ad simultaneous ac-tivation of β-AR.
