摘  要:目的建立研究气管干细胞增殖分化的动物模型。方法大鼠气管,平胸骨上缘处离断,近端应用氟尿嘧啶(5-Fu)作用30min,动态观察修复过程。结果5-Fu作用30min后,光镜下可见气管上皮细胞绝大部分脱落,仅剩余少量间隔分布的类似裸核的细胞位于基底膜上,PCNA染色阴性证实这些细胞为G0期细胞,ABCG2免疫荧光染色大部分细胞为阳性;去除5-Fu3~6h后,上皮细胞形态变为扁平;9~12h后细胞变为立方,细胞数目逐渐增多;24h上皮细胞数更多,连接成片,可见纤毛;48h接近恢复假复层纤毛柱状上皮。结论5-Fu诱发气管上皮损伤后,剩余的G0期细胞中含有气管干细胞,通过这些干细胞的增殖分化使气管上皮得以修复。[Objective] To establish the animal model of tracheal stem cells' proliferation and differentiation. [Method] The trachea of rat was cut at the level of suprasternal notch. The proximal part was injured by 5-Fu. The injury-repair process was dynamically observed. [Result] Most epithelial cells exfoliated after 30 minutes' treatment by 5-Fu. A few nail-like cells which looked like naked nuclei were randomly distributed on the basement membrane. PCNA negative staining suggested that these were Go phase cells. And most of the cells in Go phase were positive stained by ABCG2 which was considered as the marker of stem cells. After 5-Fu being removed, the number of the cells on the basal membrane increased gradually. The cells which undergoing the shape of flat and columnar differentiated into the ciliated, mucosal and basal cells. Finally, the tracheal epithelium nearly rehabilitated to pseudostratified cillated columnar epithelium 48 hours later. [Conclusion] There are tracheal stem cells among the left GO phase cells after being injured by 5-Fu. Just through the proliferation and differentiation of these cells that made the tracheal epithelia recovered.
