摘 要:目的 研究甜菜碱对大鼠酒精性脂肪肝的影响。方法 制备酒精性脂肪肝大鼠动物模型,用甜菜碱0.2~0.8g/kg干预,6周后观察血清脂质、肝功能生化指标和脂质过氧化指标的变化,并观察肝组织学改变。结果 酒精性脂肪肝大鼠给予甜菜碱0.2~0.8g/kg治疗6周后,可剂量依赖性地降低血清总胆固醇、三酰甘油、低密度脂蛋白。胆固醇、游离脂肪酸水平和增加高密度脂蛋白-胆固醇含量(P〈0.05或P〈0.01),也能剂量依赖性地降低肝重系数、肝脂质和丙二醛含量(P〈0.05或P〈0.01),较大剂量(0.8g/kg)时还可升高肝组织中超氧化物歧化酶含量(P〈0.01)。病理检查结果显示给予甜菜碱后可使肝细胞的脂肪变性程度明显减轻(P〈0.05或P〈0.01)。结论 甜菜碱对大鼠酒精性脂肪肝有较好的治疗作用,其部分机制可能是通过抗脂质过氧化和增加脂质代谢。Objective To study the effect of betaine on alcoholic fatty liver in alcohol-fed rat. Methods The model of alcoholic fatty liver rats were produced by alcohol lavage and then treated with betaine. The changes of serum lipid, liver function and lipid peroxidation were detected and pathological changes of tissues were observed. Results After 6 weeks of betaine with 0. 2 -0. 8 g/ kg, betaine dose dependently lowered the serum total cholesterol, triglyceride, low-density lipoprotein-cholesterol, free fatty acids, liver/ weight index, the amount of hepatic lipid and malonialdehyde, and increased high-density lipoprotein-cholesterol (P 〈 0.05 or P 〈 0.01 ). Betaine in the dose of 0.8 g/kg increased the amount of superoxide dismutase in the liver tissues( P 〈 0.01 ) . Compared with the controls, pathological results showed that betaine significantly decreased the degree of steatosis( P 〈 0.05 or P 〈 0.01 ) . Conclusion Betaine shows therapeutic effect in rat alcohol-induced fatty liver. This is partly by anti-lipoperoxidation and increase of lipid metabolism.
