一羟鬼笔毒肽(PHN)的制备及其毒理研究

摘　　要：利用反相高效液相色谱技术对灰花纹鹅膏菌(Amanita fuliginea)的肽类毒素进行分离,并对保留时间为60.807 min的成分进行了纯化,纯度达到99%.通过环肽检测、紫外光谱和质谱鉴定为一羟鬼笔毒肽(PHN),其相对分子质量为772.36 Da.一羟鬼笔毒肽(PHN)能引起植物细胞坏死,对小白鼠的LD50为1 414μg.kg-1,能引起小白鼠肝脏充血,血液循环衰竭.Phalloin is isolated and prepared from Amanitafuliginea by means of reversed- phage high-performance liquid chromatography, the purities of the yield are over 99%. Phalloin were identified through the cyclopeptide determination, UV spectrograph and MS. The molecular weight is 772.36 dalton. Phalloin can cause plant cellular to be necrosis. The LD50 in white mice is LD50 = 1 414μg· kg^-1. Phalloin can cause the liver of the white mice to be hyperemia and the blood circulation crock up.
