摘  要:目的探讨5-氟尿嘧啶(5-Fu)与双嘧达莫(DPM)作用对肾癌细胞生长的影响。方法以人肾癌细胞系(RCC925)为研究对象,运用长春新硷(VLB;Velban)作为参照的有/无核苷酸体外条件培养体系来研究不同浓度5-Fu(0.6、6.0、60.0 mg/L)与DPM的作用。结果当DPM浓度不变时,不同浓度5-Fu(0.6、6.0、60.0mg/L)分别对应的肾癌细胞生长抑制率组间比较差异有统计学意义(P<0.05);当5-FU浓度不变时,不同浓度DPM(0、1.25、2.50、5.00、10.00mg/L)分别对应的肾癌细胞生长抑制率组间比较差异有统计学意义(P<0.05);而且6、60mg/L浓度的5- Fu与5mg/L DPM联合应用,与单用0.078mg/L VLB对肾癌细胞的生长抑制率比较差异有统计学意义(P<0.05)。结论5-Fu与DPM联合应用可优化肾癌治疗。Objective To investigate the inhibitory effects of 5-Fu in combination with dipyridamole on the growth of renal carcinoma. Methords Renal carcinoma cell line (Rcc925) was used to compadatively study the inhibitory rate of the growth under non-nucleoside culture condition after cultured with RPMI 1640 fetal calf serum, there was manufactured signal-ceil with 0.25% trypsin for 48 hours, then continuous cultured 24hours in different concentration of 5-Fu (0.6, 6.0, 60.0 mg/L), DPM (0, 1.25, 2.50, 5.00, 10.00 mg/L) and VLB (0.0078 、0.078,0.78 mg/L) with RPMI 1640 including 10% dialysis ox serum in final concentration. Results when the same leve concentrantion of DPM being stablity, the inhibitory rate the different concentrantion of 5-Fu kept significant difference respectively (P〈0.05) ; on the othe hand, when the same leva concentrantion of 5-Fu being stablity, the inhibitory rate the different concentrantion of DPM kept significant difference respectively (P〈0.05), and the inhibitory rate of carcinoma cells in cultured with 6, 60 mg/L in combination with 5 mg/L DMP was significant difference compared with 0.78, 0. 078 mg/L VLB in sole use (P〈0.05). Conclusion The efffect to carcinoma ceil in vitro with combination with 5-Fu and DPM was significantly more than the sole use of VLB.
