摘 要:目的 在实验室条件下观察温度、pH和培养时间对黄绿青霉菌(PCV)产毒的影响。方法 用察氏培养基在不同温度、pH和培养时间条件下培养黄绿青霉菌,提取黄绿青霉素(CIT),用高效液相色谱法检测毒素含量。结果 黄绿青霉菌在高温(30℃)和低温(8℃)条件下均可产毒,且无明显差异(产毒量为222.36~231.02μg/g培养基);但在变温条件下产毒量剧增(产毒量为532.16μg/g培养基);酸性条件可有效抑制黄绿青霉菌产毒(产毒量为161.86μg/g培养基);培养10d左右产毒量最高,此后,CIT可因氧化降解而含量减少。结论 变温条件有利于黄绿青霉菌产毒;而酸性条件可抑制产毒;黄绿青霉菌培养10d左右时产毒量最高。Objective To observe the effect of temperature, pH and time on Penicillium citreoviridin's (PCV) production of toxin in laboratory. Methods PCV was cultured in Charles culture medium in different conditions, Citreoviridin(CIT) was extracted and quantitated by HPLC. Results Quantities of CIT produced by PCV were uniform at the temperature of 30℃ and 8℃ (222.36- 231.02 μg/g medium) ; PCV produced more CIT in alterant temperature(532.16 μg/g medium; PCV produced less CIT in acid culture medium(161.86 μg/g medium) ; PCV produced the most CIT after had cultured for 10 days. Conclusion PCV's production of toxin was promoted in alterant temperature. PCV's production of toxin was restrained in acid culture medium. PCVproduced maximum CIT after 10 days culture.
