摘 要:测定了16例乳腺癌及癌旁正常组织标本的17β羟类固醇脱氢酶(17β-HSD)活性和E2水平。结果显示,癌组织17β-HSD活性较正常组织明显下降,而E2水平升高。提示该酶活性及E2水平的改变与乳腺癌有关,但二者的变化无明显相关性(r=-0.354,P>0.1)The 17β-hydroxysteroid dehydrogenase(17β-HSD)activity and concentrations of estradiol(E2)were determined in non-malignant and malignant human breast tissues. The radiolabelled estradiol was used as the substrate and the activity of 17β-HSD in 800g supernatant fraction was calculated as pmol estradiol converted to estrone/mg protein 30min-1. The substrate and product were separated with TLC method. The 17β-HSD activity was significantly lower(P<0. 001 )in malignant than that of non-malignant breast tissues, but estradiol levels were significantly higher (P<0. 001) in breast cancer. The authors failed to observe the significant correlation between enzyme activity and tissue levels of estradiol in breast cancer. These results indicate that the lower 17β-HSD activity, which altered interconversion of estrogens, and the increased estradiol concentrations in breast cancer tissues may provides estrogenic stimulus and enhance the growth and progression ofbreast cancer.
