摘  要:目的:研究六亚甲基二乙酰胺(HMBA)对急性髓性白血病细胞株HL60细胞的作用及机制。方法: MTT法检测HMBA对HL60细胞的增殖抑制作用,流式细胞仪(FCM)检测HL60细胞分化抗原CD11b和CD14抗原表达,FCM检测HMBA对HL60细胞细胞周期分布的影响,逆转录-聚合酶链反应法检测HMBA对HL-60细胞细胞周期蛋白依赖性激酶-2(CDK2)mRNA表达的影响。结果:HMBA体外可以显著抑制HL60细胞的增殖。HMBA作用后HL60被阻滞于细胞周期的G0/G1期,并向成熟粒系分化;HL60细胞的CDK2基因mRNA表达下调。结论:HMBA可能通过调节某些细胞周期相关基因的表达,将HL60细胞阻滞于G0/G1期,进而促使HL60细胞朝成熟粒系分化,从而发挥对HL60白血病细胞的治疗作用。Objective: To investigate the effect of hexamethylene bisacetamide (HMBA) on HL60 cells in vitro. Method:Inhibitory effect of HMBA to proliferation of HL60 cells was measured by MTT assay. Differentiation of HL60 cells was detected by the change of expression of CDllb and CD14 on cell surface. The changed cell cycle's distribution of HL60 cells induced by HMBA was analysed by flow cytometry. RT-PCR was conducted to study the effect of HMBA on the transcription of mRNA of cyclin-dependent kinase-2(CDK2) in HL60 cells. Result:HMBA could inhibit the proliferation of HL60 cells in vitro. HMBA could commit HL60 cells to G0/G1 phase arrest. At the same time, HMBA could induce the increased expression of CDllb of HL60 cells, which showed HL60 cells' differentiation into granular line. RT-PCR indicated that the transcription of mRNA of CDK2 in HL60 cells decreased after HMBA treatment. Conclusion: HMBA can arrest HL60 cells at G0/G1 phase through regulating certain genes that are related to cell cycle, and induce HL60 cells differentiation. HMBA can inhibit proliferation of HL60 cells, which indicates HMBA has potential of treating leukemia.
