摘 要:研究大鼠诱发肝癌与β1,4半乳糖基转移酶(β1,4GT)的相关性。用高效液相层析(HPLC)纯化了荧光标记的受体底物(Gnβ1-2Mα1-6(Gnβ1-2Mα1-3)Mβ1-4Gnβ1-2Gn-PA),用β1,4GT的荧光标记受体底物的HPLC测定方法测定了大鼠诱发肝癌中肝组织的酶活性,发现随着诱癌周数的增加,肝脏酶活性逐渐增高,在第14周癌前期时酶活性达到最高,以后有所回落,但仍保持在高于对照组的水平。同时我们也发现,在胎鼠(孕20d)及出生第一天中酶活性与癌前期相似,说明在肝组织增生期此酶非常活跃。PURPOSE This article studied the relationship between DEN-induced rat liver cancer and the β1,4GT activity.METHODS The acceptor substrate(Gnβ1-2Mα1-6(Gnβ1-2Mα1-3)Mβ1-4Gnβ1-2Gn-PA) was isolated by HPLC. An assay method on β1,4GT has been established with this fluorescence-labeled substrate.Then the fluorescent reaction product is separated by reverse-phase HPLC. This assay can be used for assaying β1,4GT in crude extract of various tissues.RESULTS In the course of hepatic carcinogenesis in rat with DEN, it is found that hepatic enzyme activity is increasing gradully with the rising of weeks.The enzyme activity is the highest in precarcinoma in the 14th week. It reduces a little after that time, but keeps a higher level in comparision with the control during carcinogenesis. It is also found that the enzyme activity in fetus and one-day-aged rat is similar with that in precancer. CONCLUSIONS All these results have been showed that enzyme activity in hepatic hyperplasia is very active.
