摘  要:目的观察1mg地塞米松(dexamethasone,DEX)玻璃体注射对ET1在实验性视网膜脱离中表达的影响。方法32只白色兔32眼,分为单纯模型组和模型加药组,每组分为4个时段,单纯模型组用50μm玻璃微管在下方6:00赤道部部位视网膜造孔并注射0.5mL生理盐水于视网膜下间隙。模型加药组在模型建立后,再注射1mgDEX于玻璃体中。术后0.5h、1h、3h、6h摘除眼球作组织学观察,以鼠抗MAPK抗体检测视网膜色素上皮(retinalpigmentepithelium,RPE)细胞表达变化,并以免疫组织化学和原位杂交的方法检测ET1的表达并抽出玻璃体,用放射免疫测定法检测ET1浓度。结果实验各组兔眼术后均发生视网膜脱离,视网膜脱离范围3:00~9:00。单纯模型组的视网膜在造模后0.5h,即有ERK表达,并进入RPE细胞核,而模型加药组则4个时间段未见ERK磷酸化进入RPE细胞核。经放射免疫测定,在单纯模型组,ET1从模型建立后3h开始检测出ET1浓度,并有逐渐上升趋势,而模型加药组,4个时间段均未测得ET1浓度;而免疫组化染色,2组4个时间段在视网膜均未检测出ET1表达;原位杂交也证实,在造模后3h,单纯模型组ET1mRNA主要表达在细胞核,6h在胞浆只有较弱的着色,而模型加药组4个时间段,在RPE层均未检测出ET1mRNA。结论1mgDEX玻璃体注射不但影响外源性ET1进入视网膜下,而且抑制RPE内源性ET1的表达。Objective To observe the effect of intravitreal injection of I nag dexamethasone(DEX) on the expression of endothelin-1 (ET-1) in experimental retinal detachment. Methods Thirty-two albino rabbits (32 eyes) were divided into simple model groups and model and DEX groups of 4 intervals respectively. They were surgically detached by injecting 0.5 mL normal saline into subretinal space by a glass-made pipette with a flat tip of 50 tan diameter, and 1 mg DEX into the vitreous body in model and DEX groups,and then were studied with mouse anti-ERK and rabbits anti-ET-1 by immunohistochemistry,and ET-1 mRNA by situ hybridization and the ET-1 concentration in vitreous body by radioimmunoassay after drawing out 0.5 mL vitreous body fluid at detachment intervals of 0.5 hour, 1 hour, 3 hours and 6 hours. Results The retinal detachment showed in all 32 eyes of experimental rabbits, the detached scope was from 3 to 9 o' clock site. The ERK was phosphorylated and entered into the cell nucleus of RPE at 0.5 hour retinal detachment in the simple model groups while there was not any phosphorylated evidence at 4 intervals in the model and DEX groups. The concentration of ET-1 in vitreous body were detected at the 3rd hour and 6th hour in the simple model groups and did not in the model and DEX groups by radioimmunoassay. ET-1-like immunoreactivity did not appeared in the simple model groups and the model and DEX groups in neuroretina and RPE from 0.5 hour to 6 hours. The ET-1 mRNA expression were mainly situated in the nucleous of RPE cells and other cells of neural retina at the 3rd hour and 6th hour in the simple model groups, and did not in the model and DEX groups. Conclusion In this study of intravitreal injection of 1 mg DEX after retina being detached, DEX not only effect on the ET-1 entrance into subretinal fluid from blood, but on its expression in retina.
