摘 要:目的探讨内皮抑素对喉鳞状细胞癌(简称喉癌)细胞(Hep-2)和人脐静脉血管内皮细胞(humanumbilicalveinendothelialcell,HUVEC)的作用及其可能的机制。方法①MTT法检测,10μg/ml~50μg/ml的内皮抑素作用72小时和30μg/ml的内皮抑素作用24~72小时对Hep-2和HUVEC的影响;②电镜观察,加药前后Hep-2和HUVEC的超微结构;③RT-PCR法检测,加药前后Hep-2中survivin基因的表达;④光镜观察,内皮抑素(30μg/ml)对体外人造血管模型的影响。结果①内皮抑素(20μg/ml~50μg/ml)能抑制Hep-2和HUVEC的增殖(P<0.05,P<0.01),呈剂量-时间依赖性;②Hep-2与HUVEC药物作用组均呈凋亡改变;③RT-PCR结果,药物作用组中survivin基因表达受到部分抑制(P<0.01);④内皮抑素能抑制新生血管的形成,并能破坏新生的血管网。结论内皮抑素可能通过诱导Hep-2和HUVEC的凋亡抑制其增殖,并能破坏新生的血管。内皮抑素可能以此抑制喉癌的生长与转移。OBJECTIVE To evaluate the inhibitory effect and possibility inhibitory mechanisms of endostatin on laryngeal carcinoma cells (Hep-2) and human umbilical vein endothelial cells (HUVEC). METHODS ①Using the MTT assay , the effect of different concentrations of endostatin (10 μg/ml-50 μg/ml) and endostatin (30 μg/ml) with different contact times (24-72h) on the livability of Hep-2 and HUVEC cells were determined; ②Changes in the ultrastructures of Hep-2 and HUVEC cells were examined by electron microscopy; ③Survivin mRNA content was determined in Hep-2 cells exposed to endostatin (30 μg/ml) by RT-PCR test; ④The effect of endostatin (30 μg/ml) on the ectogenetic artificial blood vessel models was observed by light microscopy. RESULTS ①The growth of Hep-2 and HUVEC cells was significantly inhibited (P〈0.05,P〈0.01) by endostatin (20 μg/ml-50 μ/ml) in a dose-time-dependent fashion; ②Apoptosis was observed in Hep-2 and HUVEC cells exposed endostatin under the electron microscope; ③ The expression of survivin in Hep-2 cells was partly inhibited by endostatin as determined by RT-PCR (P〈0.01); ④ Endostatin not only inhibited the formation of neonatal blood vessels, but also destroyed newly formed neonatal blood vessel nets. CONCLUSION Endostatin can inhibit the proliferation of Hep-2 and HUVEC cells, possibly by inducing apoptosis and destroying neonatal blood vessels. This may be the mechanism by which endostatin inhibits the growth and the transformation of laryngeal squamaous cells carcinoma.
