摘 要:目的研究人外周血不同来源的树突状细胞(dendritic cells,DC)光镜形态学、细胞超微结构及体内外诱导抗肿瘤免疫反应的差异。方法联合应用粒/巨噬细胞集落刺激因子(GM-CSF)、白介素-4(IL-4)及肿瘤坏死因子(TNFα)从人外周血中培养出两类DC;人肝癌细胞系SMMC-7721细胞的可溶性肿瘤抗原刺激两类DC;两类DC激活同源的T淋巴细胞;观察两类DC激活的T淋巴细胞分别抑制已接种于裸鼠的人肝癌细胞系SMMC-7721移植瘤生长。结果细胞因子诱导的人外周血DC群可见到两类DC亚群:一类可能来自DC前体细胞,另一类是由单核细胞分化来的DC;两类DC体外激活的T淋巴细胞能分别抑制肝癌细胞系SMMC-7721裸鼠移植瘤生长。结论细胞因子诱导的人外周血DC群存在多相性,两类DC亚群体内外诱导的抗肿瘤细胞免疫反应不仅能抑制肝癌细胞系SMMC-7721裸鼠移植瘤发生,而且抑瘤率不同,提示两类DC亚群在肝癌的治疗中发挥着不同的作用。Objective To study diference of cytomorphological, cellular ultrastructure and the immune response in vivo and vitro induced by diverse dendritic cells(DC) pulsed with tumor extracts. Methods Isolated and purified two kind of DC from peripheral blood by combination of granulocyte/macrophage colony stimulating factor, interleukin 4 and tumor necrosis factor α; extracted tumor-associated antigen from human hepatoma cell line SMMC-7721 cells; initiated the T lymphocyte with DC pulsed by the soluble tumor antigen to produce cytotoxic T lymphocyte (CTL); tested the cytotoxic effect of CTL on SMMC-7721 cells; implanted the CTL to nudes to inhibit the growth of implanted tumor. Results Cytokines in vitro induced DC populations contained two subsets of DC, one was considered as precursor cell derived DC, The other was identified as monocyte-derived DC; The CTL induced by two kind of DC pulsed with soluble tumor antigen from SMMC-7721 cells can inhibit the growth of implanted tumor in nudes. Conclusion Cytokines in vitro induced DCs from human peripheral blood are heterogeneous,The immune response induced by two kind of DC not only inhibits the growth of implanted tumor,but also inhibits the difference of implanted tumor of SMMC-7721 cells in nudes. The results suggest, as two kind of different vaccine DC from peripheral blood pulsed by soluble tumor antigen may play different role in therapy against tumor.
