摘 要:目的:观察藤黄菌素对HL-60细胞体外增殖的抑制作用及其对细胞周期的影响。方法:分别采用台盘蓝拒染法及流式细胞术,在不同条件下测定细胞增殖的抑制率及细胞周期的分布。结果:应用台盘蓝拒染法,藤黄菌素能够显著地抑制HL-60细胞的体外增殖并且呈现时效及量效关系。流式细胞术的结果表明:藤黄菌素作用2h可使HL-60细胞的细胞周期分别阻滞于G2/M期和G0/G1期。结论:藤黄菌素能够明显抑制HL-60细胞体外增殖。Objective To observed the influence of luteolin on proliferation and cell cycle of human leukemic cell HL-60 in vitro. Methods The cell growth was tested by trypan blue stain assay and the cell cycle was analysed by flow cytometry in the various conditions. Results Using the trypan blue stain assay we found that luteolin inhibited the proliferation of HL-60 cell strongly, the higher dose and/or the longer time, the more obvious effects. Flow cytometry analysis showed that luteolin could arrest cell cycle progression in G2/M and G0/G1 phase in two hours, respectively. Conclusion luteolin is an effective flavonoid compound which can notably inhibit the proliferation and cell cycle of HL-60 cell in vitro.
