摘 要:目的:分析乳腺癌、癌前病变及正常组织中ERK和PI3-K的mRNA、蛋白表达及激酶活性,探讨其在乳腺癌发生和发展中的作用和相互关系。方法:应用RT-PCR方法检测mRNA表达;Westernblot检测蛋白表达;激酶活性检测其磷酸转移酶效率。结果:乳腺癌中ERK2mRNA和蛋白表达均增加(100%,37/37);激酶活性增加(75.68%,28/37)。有32例PI3-KmRNA表达增加(86.49%);35例蛋白表达增加(94.59%);25例激酶活性增加(67.57%)。ERK在乳腺癌和癌前病变中均处于活化状态,PI3-K只在乳腺癌中处于高表达。相关性分析表明,乳腺癌中ERK2与PI3-K激酶活性呈负相关。结论:ERK和PI3-K途径在乳腺癌发生过程中均存在异常且两者可能存在某种协调关系。To investigate the mRNA expression level, protein expression level and kinase activity of both ERK and PI3-K in breast cancer, atypical hyperplasia, benign growth.and adjacent normal mammary glands, and to discuss the correlation between them. Methods: RT-PCR, Western blot and kinase activity assay were used to detect the mRNA expression level, protein expression level and kinase activity of ERK and PI3-K, respectively. Results: For ERK, mRNA expression, protein level and kinase activity were 100%, 100% and 75.68% respectively. For PI3-K, mRNA expression, protein level and kinase activity were 86.49%, 94.59% and 67.57%, respectively. ERK was activated during the stages of both initiation and progression, while PI3-K was activated only in the progression stage of breast cancer. There was an inverse correlation between the activity of ERK2 and PI3-K in breast cancer. Conclusion: The results indicate that both ERK2 and PI3-K pathways are involved in breast cancer and may imply coordination between the two pathways in breast tumorigenesis.
