摘  要:目的研究胰岛素样生长因子-1(insulin-like growth factor-I,IGF-I)在难治性癫癎患者颞叶中的表达,探讨其对难治性癫癎的作用。方法收集难治性癫患者术后脑组织,按手术时间的先后建立难治性癫癎脑组织库,随后在脑库中按随机化原则从中抽取30例(实验组)颞叶,正常对照组为颅脑外伤手术切除的正常颞叶组织12例,采用免疫组化、western blot测定两组的IGF-I表达并进行比较。结果免疫组化结果提示IGF-1分布于神经元和胶质细胞膜,在难治性癫癎患者颞叶中的表达比对照组明显增加。western blot的结果和免疫组化结果一致。结论IGF-1在难治性癫癎患者脑组织的表达升高可能与脑的保护机制有关,这种神经营养因子的存在有利于挽救因癫癎发作而损伤的神经元。Objective To explore the expression of IGF-1 in the temporal lobes of the epilepsy patients, and then investigate the physiopathologic role in intractable epilepsy. Methods The temporal lobes of intractable epilepsy patients were used to set up the brain tissue bank of epilepsy according the time of the operation. We selected 30 temporal lobes at random and tested the expression of IGF-1 by immunohistochemistry and western blot. Nonepileptogenic control brain tissues were used for comparison. Results In this study, the results of the immunohistochemistry showed that the expression of IGF-1 were mainly in the cell membrame of neuron and gliocyte. At the same time, it was upregulated in the intractable epilepsy. The results of western blot were coincidence well with the data of immunohistochemistry. Conclusions Our study suggested that upregulation of IGF-1 might be relevant to the brain protective effect. This neurotrophic factor might have an important role in saving the dying neuron caused by the recurrent seizures.
