摘 要:目的采用低密度cDNA表达阵列研究人不同肿瘤细胞中的糖基转移酶基因家族的表达谱差异。方法TOPO-TA克隆获得硫酸基(磺基)转移酶、半乳糖基转移酶和N-乙酰氨基葡萄糖转移酶三种糖基转移酶基因家族的cDNA文库,PCR获得目的基因,逆转录标记不同肿瘤细胞株的cDNA探针,化学发光法表达阵列对人不同肿瘤细胞进行表达谱分析。结果建立了硫酸基(磺基)转移酶、半乳糖基转移酶和N-乙酰氨基葡萄糖转移酶三种糖基转移酶家族的cDNA文库,并制成低密度cDNA表达阵列。通过阵列分析发现:在不同肿瘤细胞中,除SHG44和PC12等神经细胞未检出I-GlcNAc6ST表达外,其余几种肺癌、胃癌和白血病肿瘤细胞株中均有高水平的表达;其它糖基转移酶在不同的肿瘤细胞中也存在差异表达,与目前文献报道的肿瘤细胞的糖基化特征基本一致。结论在不同肿瘤细胞中,糖基转移酶基因家族表达谱有差异,特别是磺基转移酶I-GlcNAc6ST高水平表达更值得关注,其表达差异可能与肿瘤的发生、发展有着一定的相关性。Objective To analyze the differences of glycosyhransferase gene expression profiles in different human tumor cells with cDNA arrays. Methods PCR, TOPO-TA cloning , molecular hybridization techniques and corresponding detecting system were used to evaluate the differential gene expression profiles of GlcNAc6ST, GaIT and GnT in different tumor cell lines. Results We successfully established cDNA libraries of Glc-NAc6ST, GaIT and GnT and created low-density cDNA expression profile arrays. Using these arrays we found the I-GlcNAc6ST was highly, exclusively expressed in human lungs, gastric and leukemia cancer cells. Other glycosyhransferase genes also had different expression levels in different human tumor cells, and they were all corresponed to the reports by other pieces of research. Conclusions GlcNAc6ST, GaiT and GnT have different gene expression profiles in different tumor cells, especially the high expression level of I-GlcNAc6ST was worth to do some research much deeper. There may be some relationships among the different gene expression profiles, tumorigenesis and progression.
