表皮生长因子治疗碘酸钠诱导视网膜色素上皮变性扫描电镜观察

摘　　要：通过扫描电镜观察碘酸钠对家兔视网膜色素上皮(RPE)细胞的破坏及表皮生长因子(EGF)对其影响。参照 Cohen 法从胎儿组织中提取人表皮生长因子(hEGF);给家兔静脉注入碘酸钠,分别间隔48小时和72小时再注射一次;第7天取出部分眼球,扫描电镜观察 RPE 细胞外形及数量,余右眼玻璃体内注入hEGF 提取液,左眼注等量生理盐水;7天后,取出眼球观察。结果间隔48小时组,RPE 细胞损伤较重,hEGF对其影响不大;间隔72小时组,RPE 细胞损伤较轻,hEGF 治疗组残余 RPE 细胞较多。表明 hEGF 对损伤程度较轻的 RPE 细胞可能有促修复作用。The purpose of this study is to investigate the changes of shape and number of RPE cells damaged by sodium iodate before and after the treatment with hEGF.We extracted hEGF from fetal submaxillary glands, duodenums and pancreas with Cohen's methods.The sodium iodate solution was injected intravenously into the rabbits,and at intervals of 48h or 72h,injected again.On the 7th day,the eyes of some animals were taken out to investigate the shapes and the number of the RPE cells by scanning electron microscopy.In remainder animals,hEGF was injected vitreously into the right eyes(normal saline into the left eyes in the same way). After 7 days,the shapes and the number of the RPE cells were examined.The results:(1)In group at interval of 48h,RPE ceils were damaged seriously.After the treatment with hEGF there was no difference between the experimental eyes and the control ones;(2)In group at interval of 72h,RPE cells were damaged slightly. After the treatment with hEGF the number of normal RPE cells near optie discs in experimental eyes were much more than that in control eyes.The results suggest that hEGF can improve the repair of RPE cells which were damaged slightly.
