摘 要:目的:研究β2糖蛋白I(β22GPI)与 SMMC-7721肝细胞膜的相互作用过程,探讨乙型肝炎病毒(HBV)嗜肝性的发生机制.方法:采用荧光显微镜及流式细胞仪分析的方法.检测SMMC-7721、HL-60及SGC-7901 三种细胞膜与FITC-β2GP I相结合的情况以及二者的结合特性.结果:仅SMMC-7721细胞膜表面附有大量荧光,其余2种细胞无此现象.加FITC-β2GP I 20 μL时,SMMC-772 1与FITC-β2GP I 的结合率为19%,明显高于FITC-β2GP I与 HL-60的结合率(1.7%)及与SGC-7901的结合率(1.9%)(P<0.01).加FITC-β2GP I 50μL 时,SMMC-7721与FITC-β2GP I的结合率为 20.8%,与20 μL的结合率无差异(P>0.05),说明SMMC-7721与FITC-β2GP I的结合率不随 FITC-β2GP I量的增加而增高.结论:在SMMC-7721肝细胞膜表面存在能与人β2GP I特异结合的蛋白.AIM: To further study the interaction between beta-2-glycoprotein Ⅰ (β2GPI) and the membrane of hepatoma SMMC-7721 cells, and to explore the mechanism of hepatitis B virus (HBV) infection. ETHODS: Fluorescence microscopy and fluorescence activated cell sorter (FACS) was used to observe the interaction of β2GP Ⅰ with the hepatoma cell line SMMC-7721, gastric carcinoma cell line SGC-7901 and lymphoma cell line HL-60. RESULTS: Fluorescence microscopy revealed specific binding of FITC-β2GP I to SMMC-7721 cells, but neither to HL-60 nor SGC-7901 cells. FACS analysis demonstrated that the binding rate of FITC-β2GP Ⅰ (20 μL) to SMMC-7721 cellswas significantly higher than that in HL-60 or SGC-7901 cells (19% vs 1.7%, 1.9%, both P 〈 0.01). When 50 μL FITC-β2GP Ⅰ was used, the binding rate of FITC-β2GP Ⅰ to SMMC-7721 cells was 20.8%, which was not significantly higher than 19%. The binding rate to SMMC-7721 cells did not increase with the increasing amounts of FITC-β2GP Ⅰ. CONCLUSION: There exists the specific β2GP Ⅰ binding protein in the membrane of SMMC-7721 cells.
