摘  要:目的探讨骨化三醇对大鼠局灶性脑缺血-再灌注模型的保护效应及其机制。方法将54只健康雄性成年SD大鼠随机分为三组:假手术组(8只)、骨化三醇干预组(23只,2μg.k-g1.d-1,持续8 d)和安慰剂组(23只,脂肪乳剂2m l.kg-1.d-1,持续8 d)。在第8天给药结束后1 h,以线栓法制作大鼠右侧大脑中动脉缺血后1 h再灌注模型,检测其3、6、9 h新皮质核转录因子(NF-κB)、肿瘤坏死因子(TNF)-α、白细胞介素(IL)-10及72 h时脑梗死体积及神经细胞损伤情况。结果骨化三醇干预组与安慰剂组相比,脑缺血-再灌注后3、6、9 h的NF-κB活性、TNF-α含量亦显著降低(P<0.05),6 h的IL-10含量显著增加(P<0.05)。脑缺血-再灌注72 h骨化三醇组大鼠相对脑梗死体积为(11.8±3.6)%,明显小于安慰剂组的(21.6±4.2)%,P<0.05。结论骨化三醇可能通过对炎症介质的免疫调节减轻大鼠脑缺血-再灌注性损伤。Objective To explore the protective effect of calcitriol intervention on cerebral ischemia/reperfusion (I/R) model in rats and its mechanism. Methods Fifty-four healthy adult male Spragne-Dawley rats were randomly allocated into sham-operation ( n = 8 ), calcitriol intervention ( n = 23. 2μg·kg^-1·d^-1, 8 d) and placebo groups (n =23, fat milk 2 mL·kg^-1·d^-1, 8 d). A rat model of cerebral I/R was established by the fight middle cerebral artery occlusion with the suture method on day 8, 1 hour after the administration. Neocortex nuclear factor-κB ( NF-κB), tumor necrosis factor-α (TNF-α), interleukin-10 (IL-10) at 3, 6 and 9 h after I/R, and the size of cerebral infarction and neuron injury 72 hours after I/R were measured. Resets The NF-κB activity in the calcitriol intervention group decreased significantly as compared with that in the placebo group at 3,6 and 9 h after I/R (P 〈0.05). The TNF-ct content was also decreased significantly (P 〈 0. 05 ). The IL-10 content increased significantly ( P 〈 0. 05 ) at 6 hours after I/R. The size of cerebral infarction ( 11.8 ±3. 6) % in the ealcitriol intervention group was remarkably smaller than that (21.6 ±4. 2) % in the placebo group. Conclusion The preadministration of calcitriol may reduce cerebral I/R injury in rats through the effects of immunomodulation on in- flammatory media.
