摘 要:目的:用腺病毒介导的反义VEGF165(AdaVEGF165)转染人血管内皮细胞系(ECV304)和人皮肤恶性黑色素瘤细胞系(A375)后,观察AdaVEGF165对这两细胞系体外生长的影响。方法:用感染复数为100的AdaVEGF165分别转染上述细胞,测定其生长曲线(MTT法)、细胞周期、VEGF基因的表达。结果:转染AdaVEGF165基因的A375细胞上清能够抑制ECV304细胞的生长,其增殖指数下降。AdaVEGF165对A375细胞的生长、增殖指数无明显影响,但是它能抑制A375细胞VEGF的表达。结论:①ECV304细胞不表达VEGF,AdVEGF165对ECV304细胞无直接影响,必须要经过A375细胞介导才能抑制ECV304细胞的生长;②在体外,AdaVEGF165不能直接抑制A375细胞的生长,但能抑制A375细胞分泌VEGF。Aim: To investigate the effect of Ad-aVEGF165 transfection on ECV304 and A375 cells. Methods: Ad- aVEGF165 with MOI of 100 was transfected into A375 cells and ECV304 cells. The effects of Ad-aVEGF165 on human ECV304 and A375 cells were analyzed by MTT assay, cell cycle, and the expression level of endogenous VEGF was detected using immunohistochemistry. Results: After being transfected Ad-aVEGF165 ,the A375 cell culture supernatant fluid could inhibit ECV304 cell growth markedly and decrease its proliferation index. Ad-aVEGF had no obvious effects on A375 cell growth and proliferation index, however, the down-regulation of endogenous VEGF was observed after Ad-aVEGF infection. Conclusion: ECV304 cell does not express VEGF. Ad-VEGF165 has no direct effects on ECV304 cell. With the media of A375 cell, Ad-VEGF165 can inhibit the growth of ECV304 cell. In vitro,Ad-aVEGF165 does not inhibit the growth of A375 cells, but inhibit VEGF secretion of A375 cells.
