摘 要:目的:研究视网膜母细胞瘤细胞株VEGFR-3的表达,探讨视网膜母细胞瘤自身增殖的机制。方法:选择Rb50细胞株,通过S-P免疫组化法、RT-PCR法及PCR产物琼脂糖凝胶电泳等方法,研究Rb50细胞株VEGFR-3的表达情况。结果:S-P免疫组化法呈阳性结果,表明Rb50细胞株有VEGFR-3的表达,RT-PCR产物琼脂糖凝胶电泳表明Rb50细胞株有较高水平VEGFR-3 mRNA的存在。结论:视网膜母细胞瘤细胞株有VEGFR-3的表达,表明VEGFR信息通路对视网膜母细胞瘤生长、浸润的影响,除了与促进肿瘤血管及淋巴管新生,改善肿瘤组织微环境相关外,还与促进肿瘤细胞自身增殖有关。Objective:To study the expression of VEGFR-3 in Rb50 cell strain and the mechanism of self-proliferation in retinoblastoma. Methods: Selected Rb50 cell strains were examined for expression of VEGFR-3 using S-P immunohistochemistry, RT-PCR and agarose gel electrophoresis (AGE). Results: S-P immunohistochemistry demonstrated strong VEGFR-3 expression in Rb50 cells, and AGE analysis of RT-PCR products suggested high level of VEGFR-3 mRNA in Rb50 cell strains. Conclusion: the strong expression of VEGFR-3 in Rb50 cells indicated that VEGFR-3 signal pathway may influence the development and infiltration of retinoblastoma not only by improved tumor microenvironment due to increased vascular or lymphatic neogenesis, but also through enhanced tumor selfproliferation.
