摘  要:目的探讨阿扑吗啡是否对6-羟基多巴胺毁损纹状体帕金森病鼠模型有神经保护作用。方法6-羟基多巴胺毁损大鼠左侧纹状体,在毁损前15 min阿扑吗啡(10 mg/kg,皮下注射),连续注射11 d。毁损2周后,分别进行行为学(苯丙胺引起的旋转数目)和神经化学(高压液相测定纹状体多巴胺及代谢物含量)的研究。结果阿扑吗啡能降低苯丙胺引起的向损伤侧旋转的数目。而且,显著减低多巴胺的损耗使其恢复到正常,并使DOPAC/DA比率恢复到正常。结论在6-羟基多巴胺毁损纹状体模型中,阿扑吗啡不仅改善运动功能,而且,恢复纹状体的多巴胺含量。Objective To investigate the possible neuroprotective effect of dopamine (DA) receptor agonist R- apomorphine (R- APO) on the 6 - hydroxydopamine (6 - OHDA) - induced rat model of Parkinson' s disease. Methods The rats received an injection of 6 - hydroxydopamine (6 - OHDA) into the left striatum. R - APO administration ( 10 mg/kg, s.c. ) started 15 min before 6 - OHDA injection and was repeated daily for 11 days post- injury to striatum by the injection. Behavioral test was carried out 2 weeks post - injury. Motor impairment was evaluated by amphetamine- induced rotation (5 mg/kg i.p). The degree of the injury was determined post- mortem by determination of DA and DOPAC contents in the striatum. Results R- APO treatment attenuated the amphetamine- induced ipsiversive rotation and significantly alleviated the 6 - OHDA - induced striatal DA depletion and DOPAC/DA ratios were normalized. Conclusion In the rat model of Parkinson's disease, R- APO not only significantly improves motor function but also restores DA content in the striatum.
