摘 要:目的:研究中药复方肝癌宁对人肝癌细胞体内、外生长及其对甲胎蛋白(AFP)分泌的影响.方法:以小鼠移植人肝癌模型观察药物(每天10,5,2.5 g/kg)的体内抗肝癌作用.以MTT法测定复方肝癌宁(45 g/L)的体外抗肝癌活性,以ELISA检测AFP分泌量.结果:10 g/kg对小鼠移植人肝癌HepG_2移植瘤的生长有明显抑制作用,抑瘤率为54.5% (P<0.01),且呈量效关系(r=10,P<0.01).与空白对照组相比,复方肝癌宁45 g/L作用4,5,6 d时体外培养的人肝癌HepG_2细胞株的生长明显抑制(7.0±1.3 vs 10.3±0.9,P<0.05;12.1±0.4 vs 21.3±0.9,P<0.01;15.2±0.3 vs 45.6±0.6,P<0.01),AFP分泌量也明显减少(39.6±1 vs 54.5±3;20.8±2 vs 45.6±4;18.5±2 vs 40.2±3:均P<0.01).结论:中药复方肝癌宁能明显的抑制肝癌HepG_2细胞生长及AFP分泌水平.AIM: To investigate the effects of composite Gan'aining (CGN) on the growth and alphafetoprotein (AFP) secretion of human hepatoma cells. METHODS: Mice bearing human hepatoma were treated with CGN (10, 5, and 2,5 g/kg per day), and the tumor growth was observed, In vitro inhibitory effect of CGN (45 g/L) was detected by MTT assay, and the secretion of AFP was measured by enzyme-linked immunosorbent assay (ELISA). RESULTS: CGN inhibited the growth of the transplanted hepatoma in a dose-dependent manner (r= 0, P〈0.01), and the tumor inhibition rate was 54.5% (P〈0.01) when 10g/kg CGN was used. In comparison with that in the controls, the growth of HepG2 cells was significantly inhibited at 4, 5 and 6 d (7.0±1.3 vs 10.3±0.9, P 〈 0.05; 12.1±0.4 vs 21.3±0.9, P〈0.01; 15.2±0.3 vs 45.6±0.6, P 〈 0.01), and the secretion of AFP was decreased (39.6±1 vs 54.5±3; 20.8±2 vs 45.6±4; 18.5±2 vs 40.2±3; all P〈0.01). CONCLUSION: CGN can inhibit the growth of hepatoma cells HepG2 in vitro and in vivo, and also decrease AFP secretion of HepG2 cells.
