摘 要:目的探讨硫酸基转移酶SULT1A1基因多态性与食管癌易感性的关系,及其与吸烟的交互作用。方法采用PCR—RFLP方法检测了163例新发食管癌患者和166例对照SULT1A1基因多态性,并应用多因素logistic回归等方法分析基因型对食管癌的危险度。结果①SULT1A1Arg/Arg、Arg/His、His/His三种基因型分布在病例与对照组之间差异无显著意义(P=0.60),病例与对照His等位基因分别为17.79%和12.57%,差异有显著性(P=0.028),His等位基因可显著增加患食管癌的危险性(OR调整=1.868,95%CI为1.107~3.153);②调整混杂因素后,吸烟与SULT1A1存在交互作用(OR=2.802,95%CI为1.257~6.246);③携带有His等位基因的重度吸烟者患食管癌的危险性,是不携带His等位基因的非吸烟者的8.538倍。结论SULT1A1基因突变基因型是食管癌的易感基因,它与吸烟存在联合作用。Objective To study the correlation of polymorphisms of SULT1A1 with the risk of esophageal cancer and smoke-gene interaction. Methods 163 cases of esophageal cancer and 166 controls were collected from Heping hospital and Heji hospital of Changzhi in Shanxi Province. Blood sample were collected for detecting SULT1A1 polymorphisms by PCR and RFLP. Results ① There was no significant difference in genotype of Arg/Arg, Arg/His and His/His of SULTIA1 between cases and controls ( P = 0.60) ; The frequencies of His allele were 17.79 % and 12.57 % in cases and controls, respectively (P〈0.05); the risk was significantly higher in people with His allele (OR,adjusted= 1. 868, 95 % CI was 1. 107- 3. 153). ②There was an interaction existed between smoking and the variant SULT1A1 after adjusting some confounding factors ( OR = 2. 802,95% CI was 1. 257 - 6. 246).③Heavy smokers with His allele had 8. 538-fold higher risk (95 % CI was 1. 633-44. 624). Conclusious A genetic polymorphism in the SULT1A1 gene may be associated with increased esophageal cancer, and an interactionexisted between smoking and the variant SULT1A1.
