酚妥拉明对吗啡戒断大鼠血清和下丘脑NO含量和NOS活力的影响

摘  要:目的:探讨酚妥拉明对吗啡戒断大鼠血清及下丘脑一氧化氮(NO)含量和一氧化氮合酶(NOS)活力的影响。方法:大鼠皮下注射递增剂量吗啡,建立吗啡依赖动物模型。在自然戒断症状最明显时,侧脑室注射酚妥拉明,测定血清和下丘脑匀浆NO含量和NOS活力。结果:吗啡戒断大鼠血清NO含量和NOS活力显著升高,下丘脑NO含量和NOS活力无显著变化。侧脑室注射酚妥拉明可使吗啡戒断大鼠血清NO含量和NOS活力明显下降,而对下丘脑NO含量和NOS活力无显著影响。结论:NO/NOS系统可能参与吗啡戒断,并且受α1肾上腺素能神经系统影响。Objective : To determine the effects of phentolamine on the nitric oxide (NO) contents and the nitric oxide synthase (NOS) activities in serum and hypothalamus during morphine withdrawal in rats. Methods: A physical dependent model in rats was established by subcutaneous injection of morphine. Phentolamine was injected intracerebroventricularly during the most obvious natural withdrawal syndromes. The contents of NO and the activities of NOS in serum and brain homogenate were measur. Resuits: The contents of NO and the activities of NOS increased significantly in serum, but did not chang in hypothalamus during morphine withdrawal. Phentolamine decreased the contents of NO and the activities of NOS of morphine withdrawal rats in serum, but not in hypothalamus. Conclusion : Peripheral NO/NOS system could be involved in morphine withdrawal, which might be partially mediated by α1 noradrenergic system.
