摘  要:目的探讨外源性Fas基因介导舌鳞癌细胞凋亡的可行性及其可能的分子机制。方法脂质体法将重组人Fas真核表达载体转染人舌鳞癌细胞系Tca8113,抗Fas单克隆抗体诱导其凋亡,Westernblotting检测转染前后Tca8113细胞Fas蛋白表达水平,TUNEL法检测转染前后细胞凋亡水平。结果转染后Tca8113细胞Fas蛋白表达上调;抗Fas单克隆抗体可诱导Tca8113细胞凋亡,凋亡指数升高。结论上调Fas基因表达及应用抗Fas单克隆抗体可诱导肿瘤细胞凋亡,有可能作为肿瘤基因治疗的新途径。Objective To investigate the possibility and possible mechanism of apoptosis in tongue squamous cell carcinoma mediated by exogenie Fas gene transfer. Methods Eukaryotic expressing plasmid BK-Fas was reconstructed and transferred into human tongue squamous cell eareinoma cell line Tca8113 by lipofeetamine; anti-Fas monoelonal antibody was used to induee Tca8113 apoptasis, Western blotting assay was applied to detect the expression of Fas protein pre- and post transfection; TUNEL was applied to examine the change of tumor cell apoptasis. Results Up-regulations of F as protein expression were found in Fas transfected eels. Anti-Fas monoelonal antibody could be used in the induction of tumor cell apoptasis. Conclusion Up-regulation of Fas expression can be a new pathway of tumor gene therapy to mediate tumor cell apoptosis.
