摘 要:目的比较外周血γδT细胞和αβT细胞对胰腺癌细胞株的杀伤作用,研究γδT细胞在胰腺癌过继免疫治疗中的作用。方法从胰腺癌病人外周血中分离出淋巴细胞,利用固相抗体包被方法体外特异性扩增γδT和αβT细胞,用流式细胞仪检测效应细胞的纯度,按一定的靶效比将效应细胞和胰腺癌细胞株体外混合培养。利用 MTT 法测定效应细胞的细胞毒作用。结果利用抗体包被方法可有效扩增γδT和αβT细胞,并达到较高纯度。外周血γδT细胞体外杀伤胰腺癌细胞株的作用与效应细胞纯度和靶效比成正相关,并对多株胰腺癌细胞株敏感;外周血γδT细胞的杀伤效果明显优于αβT细胞。结论外周血γδT细胞可以弥补传统效应细胞的不足,表现出较强的肿瘤免疫杀伤作用。Objective To study the potential role of γδT lymphocytes in cytotoxicity against pancreatic carcinoma cells. Methods The peripheral blood γδT and αβT cells were isolated from the patients with pancreatic carcinoma by Dicoll-Hypaque density gradient centrifugation and expanded using solid phase antibody in presence of IL-2 ex vivo. Then their purity was determined by flow cytometry and cytotoxicity examined by MTT method. Results γδT cells showed more remarkable cytolysis than aαβT cells. Meanwhile, the cytotoxic effects of γδT cells on pancreatic carcinoma cell lines depended on αβT cells' purity and target cells s/effector cell ratio (T/E ratio). The higher purity of γδT cells, the more marked cytolysis of γδT cells. Two effective T/E ratio of 1 : 5 and 1:20 were obtained using Student-Newan-Keuls test. There was no significant difference in sensitivity to γδT cells between pancreatic carcinoma cell lines and daudi. Conclusions γδT cells are cytotoxic to pancreatic carcinoma cell lines and might have a good prospect in immunotherapy of pancreatic carcinoma.
