摘  要:【目的】检测垂体腺瘤组织中的血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bF-GF)和内皮抑素mRNA的表达,探讨其与垂体腺瘤血管生成的关系及临床意义。【方法】应用逆转录PCR(RT-PCR)方法检测30例垂体腺瘤组织标本中VEGF、bFGF和内皮抑素mRNA的表达,以Ⅷ因子相关抗原(FⅧ-Rag)血管内皮细胞标记法作为MVD计数的方法,分析上述血管生成相关因子与垂体腺瘤血管生成的关系。【结果】VEGF、bFGF mRNA表达与MVD之间呈显著正相关(r=0.36,r=0.45,均P<0.05),内皮抑素表达和MVD之间无显著相关性(r=-0.17,P>0.05)。【结论】垂体腺瘤血管生成与促血管生成因子VEGF、bFGF基因水平表达的上调有关。[Objective]To detect mRNA expression of vascular endothelial growth factor (VEGF), basic fibroblast growth factor(bFGF) and endostatin in pituitary adenomas; to explore the relationship between VEGF, bFGF, endostatin mRNA expression and microvessel densilty (MVD) of pituitary adenomas. [Methods] VEGF, bFGF and endostatin mRNA expressions were determined in 30 pituitary adenomas by comparative reverse transcription-polymerase chain reaction (RT-PCR). MVD was assayed by immunohistochemical staining on factorⅧ related antigen (FⅧ-Rag)to highlight endothelial cells in microvessel . The correlations between mRNA expression of VEGF, bFGF, endostatin and MVD were studied. [Results]The mRNA expression of VEGF and bFGF positively correlated with MVD in pituitary adenomas ( P〈0.05). Significant correlation was not found between endostatin mRNA expression and MVD. [Conclusion]Up-regulation of VEGF and bFGF expression at the level of gene may play important roles and endostatin may not be critical in the angiogenesis of pituitary adenoma.
