摘 要:观察牛磺酸对心肌细胞感染 CoxsackieB3病毒(CVB3)后 Ca2+内流的影响。取新生 SD大鼠心室肌制备培养搏动心肌细胞, 18 h后接种 100 TCID50的 CVB3作为实验性病毒性心肌炎模型,并采用放射性同位素45Ca2+示踪技术,观察了 1~20 mmol/ L牛磺酸对正常及感染 CVB3后心肌细胞 Ca2+内流的影响。结果:(l)10~20 mmol/ L牛磺酸对正常心肌细胞不同胞外 Ca2+浓度([Ca2+]0, 0.5、 1.5和 4.0 mmol/ L)下的 Ca2+内流均有抑制作用,而 1mmol/ L牛磺酸只抑制高[Ca2+]0(4.0 mmol/ L)时 Ca2+内流,对低[Ca2+]0(0.5 mmol/ L)和正常[Ca2+]0(1.5 mmol/ L)时 Ca2+内流无明显作用;(2) l~20 mmol/ Lβ-丙氨酸对 Ca2+内流无明显影响;(3)感染CVB3后Ca2+内流增加,加用1mmol/ L牛磺酸后能减少感染CVB3后的Ca2+内流。结论:牛磺酸具有抑制培养心肌细胞感染CVB3后Ca2+内流的作用,这可能是牛磺酸能减轻CVB3所致细胞损伤的机理之一。PURPOSE To investigate the effects of taurine on Ca2+ influx in rat cultured cardiomyocytes after coxsackie virus B3 (CVB3) infection. METHODS Monolayer culture of spontaneously contracting rat heart cells were obtained from newborn SD rats and seeded into culture plates. 100 TCID50 of CVB3 (Nancy strain) was added as the infected group and the effect of l-20 mmol· L-l of taurine and β-alanine, a carboxylic analog of taurine, on Ca2+ influx were detected using radioactive isotope (45Ca2+) tracing techniques. RESULTS (l) 10-20 mmol· L-l of taurine have an inhibition effect on Ca2+ influx in different extracellular calcium concentrations ([Ca2+]0, i.e.: 0.5 mmol/ L, l.5 mmol/ L and 4.0 mmol/ L), I mmol· L-l of taurine had the same effect only when [Ca2+], was at 4.5 mmol/ L and without any effect when [Ca2+], was at l.5 mmol/ L or 0.5 mmol/ L; (2) 1-20 mmol/ L β-alanine have no effect on Ca2+ influx; (3) 1 mmol· L-l of taurine abated significantly the enhancement of Ca2+ influx caused by CVB3. CONCLUSIONS The present results that taurine could inhibit the increased Ca2+ influx after CVB3 attack may be one of the mechanisms of taurine attenuating the cell injury induced by CVB3 infection and probably provide a laboratory support for the clinical use of taurine in viral myocarditis.
