摘 要:目的:探讨罗勒多糖对荷NuTu-19卵巢癌大鼠模型的作用。方法:建立荷NuTu-19卵巢癌大鼠模型,体内观察罗勒多糖对荷瘤大鼠肿瘤生长及转移行为的影响,取大网膜瘤组织,提取总RNA,经逆转录、荧光标记后与R 20 s大鼠表达谱芯片杂交,经Gene P ix Pro 3.0图像处理软件分析。同时取肿瘤组织,进行常规病理学检测。结果:罗勒多糖处理组肿瘤的数量明显少于对照组,肿瘤体积小于、腹水量显著少于对照组(P<0.05);腹腔脏器转移程度积分与对照组有显著差异(P<0.01);同时采用表达谱芯片技术从药物组肿瘤组织中比较筛选出了268条差异表达基因。结论:罗勒多糖可能是一种抗卵巢癌转移的新型中草药,它对卵巢癌显现的独特效果可能是多个药效靶点综合作用的结果,本研究也显示了表达谱芯片技术在药物效应靶点研究中的优势和价值。AIM: To study the effect of basil polysaccharide on the rat model bearing NuTu - 19 cells. METH- ODS: The rat model bearing NuTu - 19 cells was treated with basil polysaccharide. The changes of the tumor cell biology and histopathology were observed. Total RNA was isolated from the tumor tissue. Hybridization assay was performed using gene expression microarray. The results were analyzed by photo disposal software - GenePix Pro3.0. RESULTS : Compared with control, the amounts of ascites in the rats treated with basil polysaccharide were decreased ( P 〈 0. 05 ) and the scores of abdominal caroinoma metastasis were changed significantly (P 〈0. 01 ). 268 differentially expressed genes were found in the tumor specimen from the rats exposed to basil polysaccharide. CONCLUSION: Basil polysaccharide may be a new anti - tumor drug and it has multi - targets. Gene expression microarray has huge advantage and value in the study of the drug target.
