摘 要:采用气相色谱法研究了淡紫拟青霉对有机磷农药辛硫磷的降解作用,并建立了从真菌菌液中检测辛硫磷残留的分析方法。结果表明,添加浓度为400μg·mL-1的辛硫磷经过淡紫拟青霉菌5d降解,降解率达100%。采用石油醚/丙酮(1∶1)为提取液,置40℃,110r·min-1振荡30min,石油醚萃取,浓缩,石油醚定容,经气相色谱测定辛硫磷残留量方法,其方法添加回收率范围为93%~99%,变异系数<5%。Gas chromatography was used to determine the degradation of phoxim by Paecilomyces lalicinus, NH-PL-03 strain, and a method was established for detecting residues of phoxim from fermentation's liquid. The result showed that mixed Paecilomyces lalicinus cultured with phoxim 400 μg·mL^-1 for 5 d, the phoxim were quickly degraded, with the degradation rate being 100%, and the degradation rates decreased with the concentration increase of added phoxim. Phoxim was extracted with a mixture of petrolic/acetone ( 1:1 V)as extraction sol- vent, treated at 40 ℃, 110 r·min^-1 for 30 min, blowtryed and concentrated by N2, and determined by GC. The results of analysis indicated that the average recovery rates ranged from 93% to 99% with the variation coefficient less than 5% (n=6) and the detection limit of phoxim residue was 3 ng·mL^-1.
