摘  要:考察IPTG及乳糖对含pTrx-AEP的BL21(DE3)诱导情况,SDS-PAGE分析两种蛋白的表达情况并结合菌浓变化,探讨乳糖替代IPTG的可行性,并在有选择性压力情况下传代,考察pTrx-AEP质粒稳定性,质粒突变率维持在一个极低的水平,传代对蛋白表达的影响不显著。We investigated the rule of protein expression that under control of T7 lacUV5 promoter when IPTG and lactose were used as inducer. In virtue of SDS-PAGE analytical method and the growth of E.coli, the feasibility of lactose as inducer was discussed. Recombinant bacteria were inoculated again after 12 hours cultivation under resistive oppression, stability of pTrx-AEP was studied, the mutative rate was very low and the ability of protein expression was stable.
