外周血有核细胞中t-PA mRNA的表达与血栓性疾病的相关性研究

摘　　要：目的探讨血栓性疾病与组织型纤溶酶原激活物(t-PA)mRNA表达的关系。方法用逆转录-PCR(RT-PCR)法检测外周血有核细胞中t-PA mRNA的表达,发色底物法测定血浆t-PA的活性。结果血栓形成组及血栓前状态组外周血有核细胞中t-PA mRNA的表达均明显增强,同正常对照组比较有显著性差异(P<0.01),两组人群血浆中t-PA的活性也都明显降低,同正常对照组比较有显著性差异(P<0.01)。外周血有核细胞中t-PA mRNA的表达与血栓前状态密切相关。结论外周血有核细胞中t-PA mRNA的表达与血栓形成及血栓前状态密切相关,有可能作为血栓前状态的早期辅助监测指标。Ohjective To investigate the relation between thrombotic events and the mRNA expression of tissue-type plasminogen activator. Methods The t-PA mRNA in peripheral blood nucleared cells was detected by RT-PCR and the t-PA activity was determined by chromogenic substrate method. Resultr The mRNA expression levels of t-PA from peripheral blood nucleared cells in the thrombosis group and pre-thrombosis group were remarkable high and notable difference was shown comparing with those of the normal group（P〈0.01）. The activities of plasm t-PA in the former two groups were reduced distinctly and had notable difference（P〈 0.01 ）. The mRNA expression of t-PA from peripheral blood nucleared cell had a close correlation with pre-thrombotic state. Conclusion The mRNA expression of t-PA in the peripheral blood nucleared ceils is closely correlated with the thrombosis and pre-thrombotis, which may be an early assistant indicator of the pre-thrombotic state.
