摘  要:用液相色谱-电喷雾离子阱串联质谱(LC-ESIITMSn)联用技术研究樟柳碱在大鼠肠内菌中的代谢。以樟柳碱优化色谱及质谱条件,探讨其色谱及质谱行为规律。将樟柳碱与大鼠肠内菌体外厌氧温孵培养,并和空白样品及樟柳碱标准品进行比较,依据被测物的多级质谱数据鉴定代谢物并阐述其结构。结果在温孵液中发现了樟柳碱的2种水解代谢产物,即莨菪品和樟柳酸。该方法灵敏度高、专属性好、样品前处理简单、快速,适合于药物代谢分析。Liquid chromatography-tandem mass spectrometry (LC-MS^n) has been used to determine anisodine and its metabolites. The chromatographic and mass spectrometric conditions were optimized using anisodine standard,and the chromatographic and mass spectrometric behavior patterns of anisodine were summarized. Anisodine was co-incubated with rat intestinal flora in vitro under lack of oxygen condition. Identification and structural elucidation of the metabolites were performed by comparing their changes in molecular masses (△M),retention-times and full scan MS^n spectra with those of the parent drug and blank plasma. The results revealed that the parent drug and its hydrolyzed metabolites (scopine and α-hydroxytropic acid) were found in the incubation solution.
