摘  要:目的建立高效液相色谱法测定冬虫夏草中麦角甾醇的含量测定方法。方法将样品直接皂化,环己烷提取,用分配柱和吸附柱净化和浓缩提取液,提取液用高效液相色谱法分离测定。采用ODS-C18色谱柱,甲醇为流动相,检测波长为282 nm,流速为1.3 ml/min。结果回归方程为Y=5 811.00X+3 338.02(X:浓度μg/ml),r=0.999 7,45.6-957.6μg/ml范围内线性关系良好,平均回收率为100.9%,RDS%为2.2%。结论本法结果准确、简便可行、分析速度快、分离效果好,适用于冬虫夏草中麦角甾醇的含量测定。Objective To establish a method to determining ergosterin in Chinese Caterpillar Fungus with HPLC. Methods The sample was directly saponified, then was abstracted by eyelohexane, Extract was to be cleaned by partition column and condensed by absorption column, then separated and determined by HPLC. A column of Oil-(218 was used,consisted with methanol as the mobile phase. The flow rate was 1.3 ml/min, and the UV detection wavelength was set at 282 nm. Results The calibration curve was Y = 5 811.00X + 3 338.02( X: concentration μg/ml), r = 0,999 7. The standard curve was linear over the range of 45.6 - 957.6μg/ ml, the average recovery was 100.9 % with RSD of 2.2 %. Conclusion The method is simple and accurate, so it can be suitable for quality control of the preparation of Chinese caterpillar fungus.
