摘  要:通过对蛋黄中抗嗜水气单胞菌IgY的提取,分别进行了两种粗提方法(水稀释法、Pectin法)和纯化方法(硫酸铵法、PEG6000法)的比较。结果显示,当蛋黄液经pH5.0蒸馏水10倍稀释粗提后,上清液中脂肪和IgY残余率分别为32.69%和68.34%,效果较好。采用12%(w/v)PEG6000、33%(v/v)饱和硫酸铵纯化IgY,其免疫活性残余率分别为63.5%和64.1%。SDS-PAGE显示,水稀释法和Pectin法提取的上清液中杂蛋白减少,硫酸铵和PEG6000纯化后的上清液中几乎无其他杂带。研究表明,水稀释法和Pectin法提取IgY具有简便、高效和无污染的特点,硫酸铵和PEG6000适合纯化IgY。Different methods of extracting(water dilution and Pectin) and purifying(saturated ammonium sulfate and PEG 6 000) were conducted to get IgY against Aeromonos Hydrophila. The results showed that remaining ratio of fat and IgY were 32.69% and 68.34% in suspension of egg yolk extracted with 10-floder diluted water at pH 5.0 and the effect was better. Remaining immunocompetence ratio of IgY was 63.5% and 64.1% purified by 12% PEG 6 000(w/v) and 33% saturated ammonium sulfate (v/v) respectively. SDS-PAGE indicated the amount of non-IgY protein significantly decreased by water dilution and Pectin methods,and there were no other proteins except IgY in supernatants with PEG 6 000 and saturated ammonium sulfate as precipitants. Thin paper showed water dilution and Pectin methods provided a simple,rapid and efficient means of extracting crude IgY,and PEG 6 000 and saturated ammonium sulfate were suitable to purify IgY in suspension.
