氧诱导小鼠视网膜病变模型的研究

摘  要:目的建立对视网膜新生血管进行定量研究的小鼠模型,用于视网膜新生血管发生机制的探讨及治疗方法的评价。方法给氧组16只C57BL/6J小鼠P7到P12置于氧浓度为70%±5%的氧箱,诱导视网膜新生血管形成。对照组16只C57BL/6J小鼠生活在正常氧环境中作为对照。P17各再随机分成两组,一组FITC-Dextran灌注后,视网膜铺片,在荧光显微镜下观察视网膜血管生长情况并行视网膜病变评分,视网膜病变评分包括血管生长、血管丛、视网膜外新生血管、中央血管收缩、视网膜出血和血管扭曲;另一组PAS-苏木精染色作眼球冰冻切片,计数突破内界膜的内皮细胞数。结果给氧组视网膜血管分布及形态较对照组有明显差异,给氧组视网膜中央毛细血管闭塞,大血管管径粗细不均,周边更扭曲,血管丛增生,荧光素渗漏。给氧组视网膜病变总分用四分位数表示为10(9,10),而对照组为0.5(0,1),两组比较有显著性差异(P<0.001)。突破内界膜的内皮细胞数给氧组为58.2±9.9,而对照组小于1,两者比较有显著性差异(P<0.001)。结论高氧可诱导建立小鼠视网膜新生血管模型,其发生与缺氧有关。采用F1TC-Dextran造影铺片是观察视网膜新生血管的有效方法。Objective To establish a quantifying model of retinal neovascularization suitable for the study of pathogenesis and therapeutic intervention for the retinal neovascularization. Methods The mouse model of oxygen-induced retinopathy consisting of a 5 day exposure in (75%±5%) oxygen from P7 (postnatal day 7) to P12 was used to produced retinal neovascularization. FITC-Dextran angiography of retinal vasculture was performed and retinal whole mounts were prepared to score the features of retinopathy. The parameters that were scored included blood vessel growth, blood vessel tuff formation, extra retinal neovascularization, degree of central vasoconstriction, retinal hemorrhage, and tortuosity of vessles. In addition, quantification of retinal neovascular nuclei was performed with periodic acid Schiff (PAS) staining of frozen eye sections. Results There were marked difference of the morphology of the retinal vessles between oxygen-induced group and control group. In the oxygen-induced group, the large vessles were distored, the capillary was obstructed, and lots of neovascular turfs was proliferated in the periphery of the retina. Control group had a lower median retinopathy score of 0.5 (25th, 75th quartile: 0, 1) compared with oxygen-induced group with a median score of 10 (25th, 75th quartile= 9, 10; P〈0. 001). Control group had a significant reduction in the number of nuclei extending beyond the inner limiting membrame (〈 1) when compared with oxygen-induced group (56.2±9.9, Mean±SD; P〈0. 001) . Conclusion The quantifying model of retinal neovascularization may fascilitate the further research of pathogenesis of retinal neovascularization and medicalintervention.
