It based upon the size ratio between the cloned gene and its haploid genome. From the size fraction and the amount of genomic DNA used for analysis, the gene copy number could be estimated from the data of dot blot or Southern blot analysis. Using the procedure proposed by this study, the copy numbers of human α 1 antitrypsin gene and rat RALRS 1 repetitive sequence were determined.
